Two villains with theatrical backgrounds such as these guys are naturally going to cross paths at some point. I can only imagine what kind of trouble that they would be able to cook up together.

With a title such as this one, I can only speculate that they're targeting someone high up in the local government. Probably Commissioner Gordon. And, as Clayface is a metamorph, it's not too difficult to assume that he's going to impersonate that particular police officer!
Clayface will probably start with a Shakespeare quote !
and mysterio will steal the scene…
Maybe....assassination doesn't have to be to kill a person...how about a reputation or a project. Make it look like somebody stole, took a bribe, etc. Mysterio makes sure the real person has no alibi while Clayface is working. Could be a complicated scheme and that's why it's not easy to pull off.
Or... Things fall apart, and they try to double cross each other. Perhaps the Batman, a master of disguise himself, and maybe even Mystique will step in to steal the spotlight.
I take it this is Clayface I (Basil Karlo) and not Matt Hagen or one of the other, later incarnations? (I hadn't realized that Karlo had acquired shape-shifting powers, but a quick online search reveals that indeed he did.)
@Det. Tobor: in other words, their scheme is more liable...to be about libel.
@Carycomic: as a certain wise guy once said; "Be still, my foolish stomach."
